Nicest I've ever stayed in is the Bellagio, but that's probably old by now. I think the Wynn is pretty fancypants these days. Mostly I just stay in the Luxor because hey, giant pyramid.
yea- the Wynn gets good reviews from my friends that go to Vegas a lot too. A friend just got back from there, and I think she always goes to the Wynn. sounds great too
I had a great time at The Signature at MGM Grand. Every room is a suite that comes with microwave, real mini-fridge (not a mini bar), stove top, dishes and silverware so you can go get some cheap groceries and have cereal for breakfast (as opposed to paying for breakfast/room service), the beds are comfy (even the pull out sofa), double sinks in the bathroom, as well as a stand up shower and a HUGE whirlpool tub (we somehow crammed four people in it, though that's not recommended), it's not overrun with kids, very quiet, you have your own pools and workout rooms but are allowed to use everything at the MGM Grand facilities, only $120 or so for the cheapest rooms.
Downsides: The cheapest rooms only have 1 king sized bed and a queen-sized sleeper sofa, if that matters. Also, it's not on the strip technically. It's just off the strip. You can get to the strip walking indoors through a bunch of hotel lobbies, some people movers, the shops at the MGM Grand, then the MGM casino floor. We were in the middle tower and it took us 15 minutes to go from room to the sidewalk on the strip. For some people that's a big deal. For us, the niceness of the room more than made up for it.
we got a great deal from http://www.i4vegas.com/ for the tuscany suites. the hotel is basically like staying at your aunt's condo, but it was $25/night and the bathroom was ridiculously huge. they have deals for other places too, I like to look at tripadvisor.com for pictures guests have posted.
The Luxor is my favorite in terms of layout, nice rooms, etc., and has the tram connecting you to Mandalay Bay and Excalibur. MGM Grand, Paris, Venetian, and Aladdin (now Planet Hollywood) are all good, too.

Yeah, my best Vegas hotel will probably be in October when I also stay at The Signature at MGM Grand for the Matador 21 weekend thing.

I've stayed at the Tuscany several times. The rooms are OK -- very spacious but not fancy. They have a nice outdoor hot tub, too.
